Please explain the Densitometer

Can someone explain how I use the densitometer please. Feel free to keep it as simple as possible.
For example, If I were to place the cursor over a prescan of a white patch, the densitometer might read:
R152 -> 244
G147 ->237
B153 ->246
I assume that this patch of white should ideally read about 245,245,245. What is the densitometer telling me? And just as important, how can I make adjustments to the RGB on the left is the same as the rgb on the right. Or, do I have it all wrong?
I have a transparency that I am trying to scan and there is a patch of very dark green that always comes out black. I cannot seem to get the green to show. Is this where the densitometer is used?
