how do i scan sprocket holes with out auto ajustment?

i am trying to include the 35mm sprocket holes in my scans as i have toy camera that exposes them. every time i move the scan area to include the sprocket holes the scan settings are adjusted and the exposure is thrown off. is there any way i can select the image so that software ajusts it then turn of the auto mode so that them select the spoket holes?

you can see in the below example what happens when move the scan area to include the spoeket holes:
